Significance and Uses

Dehydroeprandrosterone (DHEA) is a steroid hormone produced by the adrenal glands, gonads, adipose tissue and the brain. DHEA is structurally similar to, and is a precursor of, testosterone and estrogen. In blood, most DHEA is found as its derivative dehydroeprandrosterone sulfate, DHEA-S. In humans, DHEA production peaks in early adulthood, around the age of 25, and declines afterwards to approximately 10% of peak levels by age 80. The significance of DHEA in health and disease is not fully established.

Assay Method

Dehydroepiandrosterone sulphate (DHEAS) is measured with an automated chemiluminescent assay using the Bayer Diagnostics ACS:180. (1) The chemiluminescent immunoassay involved the competitive binding of a Dimethylacridinium ester (DMAE) labeled DHEAS derivative to a rabbit anti-DHEAS antibody. The solid phase is Goat anti-rabbit IgG conugated to superparamagnetic particles (PMP). Closely related steroids show little cross reactivity in the assay indicating high specificity. The assay demonstrates acceptable parallelism of diluted serum samples and recovery of added exogenous hormone. DHEA cross reacts 192%, but the low levels of serum DHEA render this inconsequential. The intra assay coefficient of variation is 8.02% (n=261) and inter assay coefficient of variation is 11.34% (53.32 ug/dL, n=37) and 9.74% (250.21 ug/dL, n=37).
